目的探讨E-cadherin及Survivin在不同临床病理学类型胃癌中的表达及二者的相关性,分析它们在胃癌发生、浸润及转移中的作用,为胃癌诊治和判断预后提供依据。方法采用免疫组化法(S-P法),检测E-cadherin、Survivin在胃癌及癌旁组织中的表达,分析它们与胃癌各临床病理特征之间的关系。结果 (1)胃癌组织中E-cadherin阳性表达率低于癌旁正常黏膜组织,Survivin阳性表达率高于癌旁正常黏膜组织(P均<0.01)。(2)胃癌组织E-cadherin表达与胃癌分化程度、浸润深度、淋巴结转移及远处转移显著相关(P<0.05或0.01),Survivin表达与胃癌浸润深度、淋巴结转移、远处转移及TNM分期显著相关(P<0.05或0.01)。(3)胃癌组织中,E-cadherin和Survivin表达呈明显负相关性(r=-0.504,P<0.01)。结论 (1)E-cadherin、Survivin表达异常参与了胃癌发生发展过程,可作为检测早期胃癌的一个生物学标志。(2)E-cadherin、Survivin在胃癌中表达异常可导致胃癌细胞更具侵袭性,易于转移,可作为预测胃癌进展和预后的指标。(3)在胃癌中,E-cadherin与Survivins阳性表达呈负相关性,提示二者在胃癌的发生发展过程中可能存在一定关系。
Objective To investigate the expression of E-cadherin and Survivin in gastric cancer of different clinicopathological types and the correlation between them, analyze their roles in the occurrence, invasion and metastasis of gastric cancer and provide the basis for diagnosis and prognosis of gastric cancer. Methods The expressions of E-cadherin and Survivin in gastric cancer and adjacent normal tissues were detected by immunohistochemistry (S-P method), and their relationship with the clinicopathological characteristics of gastric cancer was analyzed. Results (1) The positive expression rate of E-cadherin in gastric cancer tissues was lower than that in normal mucosa tissues, the positive rate of Survivin expression was higher than that in normal mucosa (P <0.01). (2) There was a significant correlation between the expression of E-cadherin and the differentiation degree, depth of invasion, lymph node metastasis and distant metastasis (P <0.05 or 0.01), the expression of Survivin and the depth of gastric cancer invasion, lymph node metastasis, distant metastasis and TNM staging (P <0.05 or 0.01). (3) The expression of E-cadherin and Survivin in gastric cancer tissues was negatively correlated (r = -0.504, P <0.01). Conclusions (1) The abnormal expression of E-cadherin and Survivin is involved in the development and progression of gastric cancer, which can be used as a biomarker to detect early gastric cancer. (2) The abnormal expression of E-cadherin and Survivin in gastric cancer can lead to more invasive and metastatic gastric cancer cells, which can be used as an index to predict the progress and prognosis of gastric cancer. (3) There was a negative correlation between the expression of E-cadherin and Survivins in gastric cancer, suggesting that there may be some relationship between them in the occurrence and development of gastric cancer.
