目的:探讨Prdx1、Spy1、p27~(kip1)蛋白在肝细胞肝癌中的表达及相关性。方法:应用Western blot及免疫组化检测肝癌及相应的癌旁组织中Prdx1、Spy1、p27~(kip1)的表达;免疫共沉淀分析Prdx1、Spy1的相互作用;Western blot分析Prdx1、Spy1相互结合对p27~(kip1)蛋白表达的影响。结果:Prdx1、Spy1在肝癌组织及肝癌细胞中高表达,与p27~(kip1)表达呈负相关;Prdx1、Spy1相互结合使p27~(kip1)的表达水平下调,此作用与Prdx1的抗氧化物活性无关。结论:Prdx1、Spy1蛋白的表达与肝细胞肝癌的发生发展有关,两者的表达可能在肿瘤的恶性进展中起协同作用,而与p27~(kip1)的表达呈拮抗作用。
Objective: To investigate the expression and correlation of Prdx1, Spy1 and p27 kip1 proteins in hepatocellular carcinoma. Methods: The expressions of Prdx1, Spy1 and p27 kip1 in hepatocellular carcinoma and its corresponding paracancerous tissues were detected by Western blot and immunohistochemistry. The interaction between Prdx1 and Spy1 was analyzed by co-immunoprecipitation. The interaction between Prdx1 and Spy1 p27 ~ (kip1) protein expression. Results: Prdx1 and Spy1 were highly expressed in hepatocellular carcinoma and hepatocellular carcinoma cells, and negatively correlated with the expression of p27 kip1. The combination of Prdx1 and Spy1 reduced the expression of p27 kip1, which was related to the activity of Prdx1 Nothing to do CONCLUSION: The expression of Prdx1 and Spy1 is related to the development of hepatocellular carcinoma. The expression of Prdx1 and Spy1 may play a synergistic role in the malignant progression of tumor and antagonize the expression of p27 kip1.
