背景:一氧化氮在肿瘤的发生发展中起着双向作用,高浓度的一氧化氮通过细胞毒作用和诱导肿瘤细胞凋亡起抗肿瘤作用;在肿瘤组织改变的微环境下,其对肿瘤组织的影响主要表现为促进肿瘤的生长。目的:观察一氧化氮合酶抑制剂N-硝基精氨酸甲酯对人大肠癌细胞LS174T侵袭、运动能力的影响。设计:单一样本观察。单位:哈尔滨医科大学附属肿瘤医院外科。材料:实验于2004-01/10在哈尔滨医科大学肿瘤研究所完成,人大肠癌细胞系LS174T购自哈尔滨医科大学肿瘤研究所,N-硝基精氨酸甲酯购自Sigma公司。方法:采用0.2,0.4,0.8和1.0mmol/LN-硝基精氨酸甲酯对LS174T细胞进行干预,用重组基底膜侵袭实验观察药物对癌细胞侵袭、运动能力影响。主要观察指标:N-硝基精氨酸甲酯对LS174T细胞侵袭重建基底膜的抑制率和趋化运动抑制率。结果:①0.2,0.4,0.8和1.0mmol/LN-硝基精氨酸甲酯处理LS174T细胞72h,对细胞侵袭重建基底膜的抑制率分别为10.29%,19.62%,34.08%,42.23%(P<0.05或0.01)。②0.2,0.4,0.8和1.0mmol/LN-硝基精氨酸甲酯处理LS174T细胞72h,对细胞趋化运动抑制率分别为20.76%,24.95%,39.43%,46.85%(P<0.01)。结论:N-硝基精氨酸甲酯具有抑制LS174T细胞侵袭和转移的作用。
BACKGROUND: Nitric oxide plays a bidirectional role in the development and progression of tumors. High concentration of nitric oxide acts as an antitumor agent through cytotoxicity and induces apoptosis of tumor cells. Objective: To observe the effect of nitric oxide synthase inhibitor N-nitro-arginine methyl ester on invasion and exercise capacity of human colorectal cancer cell line LS174T. Design: Single sample observation. Unit: Affiliated Tumor Hospital, Harbin Medical University. MATERIALS: The experiment was performed at Institute of Oncology, Harbin Medical University from January to October 2004. The human colorectal cancer cell line LS174T was purchased from Institute of Oncology, Harbin Medical University. N-nitro arginine methyl ester was purchased from Sigma Company. Methods: The LS174T cells were treated with 0.2, 0.4, 0.8 and 1.0 mmol / L N-nitroarginine methyl ester, and the effect of drugs on invasion and exercise ability of cancer cells was observed by recombinant basement membrane invasion assay. MAIN OUTCOME MEASURES: The inhibitory rate and chemotactic inhibition rate of N-nitroarginine methyl ester on invasion and reconstruction of LS174T cells. Results: ① After treated with 0.2, 0.4, 0.8 and 1.0 mmol / L N-nitroarginine methyl ester for 72 h, the inhibitory rates of invasion, invasion and reconstruction of basement membrane were 10.29%, 19.62%, 34.08% and 42.23% P <0.05 or 0.01). The inhibition rates of chemotactic activity of LS174T cells treated with 0.2, 0.4, 0.8 and 1.0 mmol / L N-nitroarginine methyl ester for 72 h were 20.76%, 24.95%, 39.43% and 46.85%, respectively (P <0.01) . Conclusion: N-nitro-arginine methyl ester can inhibit the invasion and metastasis of LS174T cells.
