目的对从我国南海红树林底泥中分离的一株放线菌(No.H83-3)进行活性化学成分研究。方法对H83-3发酵物的菌丝体采用90%乙醇提取和不同极性溶剂萃取,并对具有活性的氯仿部位进行研究。整个过程采用纸碟片法进行活性追踪分离,先后通过硅胶柱、开放性ODS柱以及制备型HPLC进行化合物的分离纯化;用NMR、MS等光谱方法,结合文献鉴定化合物的结构。结果分离出6个化合物,并分别鉴定为衣霉素(tunicamycin)Ⅶ(化合物1)、衣霉素D2(化合物2)、衣霉素Ⅴ(化合物3)、衣霉素S15a(化合物4),衣霉素Ⅱ(化合物5),衣霉素U17a(化合物6)。结论化合物1~6均为首次从海洋放线菌中分离得到。
Objective To study the active chemical components of an actinomycete (No.H83-3) isolated from mangrove sediment of South China Sea. Methods The mycelium of H83-3 ferment was extracted with 90% ethanol and extracted with different polar solvents, and the active chloroform site was studied. The whole process using paper disc method for the activity of the tracing separation, successively through silica gel column, open ODS column and preparative HPLC compounds were isolated and purified; with NMR, MS and other spectroscopic methods, combined with the literature identified the structure of the compound. As a result, six compounds were isolated and identified as tunicamycin VII (compound 1), tunicamycin D2 (compound 2), tunicamycin V (compound 3), tunicamycin S15a (compound 4) Tunicamycin II (compound 5), tunicamycin U17a (compound 6). Conclusion Compounds 1 ~ 6 were isolated from marine actinomycetes for the first time.
