[目的]观察二甲基亚砜(DMSO)对体外大鼠结肠平滑肌收缩的影响,探讨其作为脂溶性中药成分助溶剂的可行浓度范围。[方法]制备SD大鼠结肠平滑肌肌条,以9 g/L NaCl空白对照(NS)组和乙酰胆碱对照(Ach)组,分别观察不同浓度DMSO对肌条自发收缩以及对Ach引起的肌条收缩的影响。[结果]DMSO在0.2%~1.4%浓度范围,对肌条自发收缩振幅和面积的影响与NS组比较均无统计学意义(P>0.05)。与Ach组比较,在0.2%~1.8%浓度范围,DMSO对Ach引起的肌条收缩振幅和面积的影响均无统计学意义(P>0.05)。[结论]DMSO在浓度≤1.4%时对体外大鼠结肠平滑肌的收缩无明显影响,作为脂溶性中药的助溶剂有运用价值。
[Objective] To observe the effect of dimethyl sulfoxide (DMSO) on the contraction of rat colonic smooth muscle in vitro and explore its feasible concentration range as a cosolvent for fat-soluble Chinese medicine. [Methods] SD rat colonic smooth muscle strips were prepared. The 9 g/L NaCl blank control (NS) group and the acetylcholine control (Ach) group were used to observe the effects of different concentrations of DMSO on the spontaneous contraction of muscle strips and the contraction of Ach-induced muscle strips. Impact. [Results] The effects of DMSO on the amplitude and area of spontaneous contraction of muscle strips were not statistically different from that of NS group (0.2%~1.4%) (P>0.05). Compared with the Ach group, in the concentration range of 0.2%~1.8%, the effect of DMSO on the amplitude and area of muscle strip contraction caused by Ach was not statistically significant (P>0.05). [Conclusion] When DMSO concentration is less than 1.4%, it has no obvious effect on contraction of rat colon smooth muscle in vitro, and it has application value as a cosolvent for fat-soluble Chinese medicine.
