目的探讨大豆黄酮对肝癌SMMC7721细胞糖代谢和癌干细胞CD133基因表达的影响。方法培养人肝癌SMMC7721细胞并用大豆黄酮处理。MTT法测定细胞的成活率,用试剂盒法测定人肝癌SMMC7721细胞中的己糖激酶活性,并用Western Blot方法测定癌干细胞CD133蛋白的水平。结果大豆黄酮对人肝癌SMMC7721细胞增殖具有抑制作用,同时,可降低人肝癌SMMC7721细胞中己糖激酶活性和癌干细胞CD133蛋白的水平。结论大豆黄酮能抑制人肝癌SMMC7721细胞的增殖,降低人肝癌SMMC7721细胞中己糖激酶活性和癌干细胞CD133蛋白的水平。
Objective To investigate the effects of daidzein on carbohydrate metabolism and CD133 gene expression in hepatoma SMMC7721 cells. Methods Human hepatoma SMMC7721 cells were cultured and treated with daidzein. The survival rate of cells was measured by MTT assay. The activity of hexokinase in human hepatoma SMMC7721 cells was assayed by kit method. The level of CD133 protein in cancer stem cells was determined by Western Blot. Results Daidzein inhibited the proliferation of human hepatocellular carcinoma SMMC7721 cells and decreased the activity of hexokinase and the level of CD133 protein in cancer stem cells of SMMC7721 cells. Conclusion Daidzein can inhibit the proliferation of human hepatoma SMMC7721 cells and decrease the activity of hexokinase and the level of CD133 protein in cancer stem cells of SMMC7721 cells.
