目的:探讨祛瘀化痰、散结消癥中药复方对肾小球系膜细胞增殖及ECM合成调控系统的影响。方法:以脂多糖为刺激因子,作用于体外培养的大鼠肾小球系膜细胞,设立空白对照组、川芎组、川芎+鳖甲组、川芎+海藻组、川芎+鳖甲+海藻组(高、中、低剂量组),以MTT比色法观察各组肾小球系膜细胞增殖情况,ELISA法观测细胞上清中FN水平的变化。结果:川芎组、川芎+鳖甲组、川芎+鳖甲+海藻(高、中剂量)组均能不同程度地抑制肾小球系膜细胞增殖,减少FN的含量,中以川芎+鳖甲+海藻组(高剂量)作用最明显,且随着用药浓度增加,作用增强。结论:祛瘀化痰、散结消癥中药复方具有干预肾小球硬化的作用,而这一作用可能与有效抑制系膜细胞增殖、下调系膜细胞上清中FN水平,减少ECM合成有关。
Objective: To investigate the effect of Chinese herbal compound prescriptions on the proliferation of glomerular mesangial cells and ECM synthesis and regulation system. The proliferation of mesangial cells in each group was observed by MTT colorimetric assay. The changes of FN in supernatants were observed by ELISA. Results: Chuanxiong group, Chuanxiong + turtle group, Chuanxiong + turtle shell + seaweed (high dose group) can inhibit the proliferation of glomerular mesangial cells to a certain extent, reduce the content of FN, Chuanxiong + turtle shell + Seaweed group (high dose) the most obvious role, and with the drug concentration increases, the role of enhanced. Conclusion: The traditional Chinese medicine recipe of Quyu Huatan and Sanjiexiaozhuo have the effect of intervention on glomerulosclerosis, and this effect may be related to the effective inhibition of mesangial cell proliferation, down-regulation of FN in mesangial cell supernatant and reduction of ECM synthesis.
