目的 :观察大豆低聚糖的肠道微生物的作用效应并建立一种大豆低聚糖各组分含量的检测方法。方法 :采用青春双歧杆菌和产气荚膜梭菌为肠道微生物指示菌 ;使用 μBondpak/CH氨基柱 ,高压液相色谱法 ,以纯水为洗脱液 ,分离大豆低聚糖各组分。结果 :大豆低聚糖能有效促进青春双歧杆菌在体外的增殖 ,而产气荚膜梭菌则几乎不能利用 ;HPLC法有效分离和准确测定大豆低聚糖中的各组份。结论 :大豆低聚糖是双歧杆菌的有效促生因子 ,HPLC方法是一种快速、准确、前处理简单、分离度高的含量检测方法。
Objective: To observe the effect of gut microbiota on soybean oligosaccharides and establish a method for the determination of the content of each component of soybean oligosaccharides. METHODS: Bifidobacterium adolescentis and Clostridium perfringens were used as intestinal microbial indicator bacteria; Soybean oligosaccharide fractions were separated using μBondpak/CH amino-column, high pressure liquid chromatography and pure water as eluent. . Results: Soybean oligosaccharides could effectively promote the proliferation of B. adolescentis in vitro, but Clostridium perfringens could hardly be used. HPLC method effectively separated and accurately determined the components of soybean oligosaccharides. Conclusion : Soybean oligosaccharide is an effective growth promoting factor for bifidobacteria. HPLC method is a rapid, accurate, simple pretreatment and high resolution assay.
