目的了解鼻咽癌肿瘤细胞增殖与凋亡的关系及bcl2表达在发病机制中的作用。方法15例癌旁组织,65例鼻咽癌用免疫组化技术标记bcl2、PCNA,HE染色中作核分裂像计数,并将后两者合并为增殖指数;用ISEL法显示44例鼻咽癌及15例癌旁组织中的细胞凋亡。结果①鼻咽癌PCNA、核分裂像、凋亡细胞数均显著高于癌旁组织,但均与组织分型无关。增殖指数与凋亡细胞数呈正相关。②bcl2在鼻咽癌中高表达,与组织分型有关,与凋亡细胞数呈负相关。结论①鼻咽癌肿瘤细胞具有高水平增殖的同时也具有高水平的凋亡,其肿瘤本身是高水平基础上增殖与凋亡失衡的结果。②bcl2不是调控肿瘤细胞凋亡的唯一因素,但其抑制细胞凋亡的功能在鼻咽癌发病机制中有重要作用。
Objective To understand the relationship between the proliferation and apoptosis of nasopharyngeal carcinoma cells and the role of bcl2 in pathogenesis. Methods Fifteen cases of paracancerous tissues and 65 cases of nasopharyngeal carcinoma were labeled with bcl2 by immunohistochemistry. The numbers of mitosis were counted by PCNA and HE staining, and the latter two were combined into proliferation index. The results of ISEL showed that 44 cases of nasopharynx Cancer and 15 cases of paracancerous tissue apoptosis. Results ① The numbers of PCNA, mitosis and apoptotic cells in nasopharyngeal carcinoma were significantly higher than those in paracancerous tissues, but none of them were related to the tissue typing. Proliferation index and apoptotic cells were positively correlated. ② bcl 2 in nasopharyngeal carcinoma, high expression, and tissue type, and the number of apoptotic cells was negatively correlated. Conclusions ① The NPC cells have high level of apoptosis as well as high level of apoptosis. The tumor itself is the result of a high level of imbalance between proliferation and apoptosis. ② bcl 2 is not the only factor regulating tumor cell apoptosis, but its function of inhibiting apoptosis plays an important role in the pathogenesis of nasopharyngeal carcinoma.
