目的确认实验用大白鼠肾损伤的致病因子为三聚氰胺、三聚氰酸,探讨三聚氰胺、三聚氰酸的肾毒性。方法清洁级Wistar大白鼠,在清洁级动物室喂饲含致病因子的饲料,30天后处死动物做血液生化检测、脏器系数计算,取肾组织常规制片,进行病理组织学检查。取包埋于石蜡块中的肾组织用液相色谱-质谱联用法分析肾组织中是否含有三聚氰胺、三聚氰酸。结果大鼠喂饲含致病因子的饲料后,大鼠肾内形成结晶体,并引起严重肾功能损伤,甚至衰竭;每克肾组织中含有三聚氰胺15微克、三聚氰酸60微克。结论肾脏为三聚氰胺、三聚氰酸毒作用的主要靶器官,大鼠经口喂饲一定量三聚氰胺、三聚氰酸,可致严重肾损伤。
Objective To verify the experimental rat kidney injury risk factor for melamine, cyanuric acid, melamine, cyanuric nephrotoxicity. Methods Wistar rats were cleaned and feedstuffs containing virulence factors were fed in a clean animal room. After 30 days, animals were sacrificed to perform blood biochemical tests and organ coefficient calculations. Conventional renal tissue samples were obtained for histopathological examination. The kidney tissue embedded in the paraffin block was analyzed for the presence of melamine and cyanuric acid in the renal tissue by liquid chromatography-mass spectrometry. Results Rats fed diets containing virulence factors formed crystals in the kidneys of rats and caused severe renal impairment or even failure. Each gram of kidneys contained 15 micrograms of melamine and 60 micrograms of cyanuric acid. Conclusion The kidney is the main target organ of melamine and cyanuric acid poisoning. Rats are fed with a certain amount of melamine and cyanuric acid orally and can cause serious renal damage.
