目的:研究碘化钾对染铅肾小管上皮细胞(HK-2)XIAP的表达及凋亡的影响,探讨其对抵抗铅导致的近曲肾小管上皮细胞损伤的可能机制。方法:应用细胞组化法检测染铅HK-2细胞及对照组细胞中XIAP的表达;流式细胞术检测其凋亡情况。结果:染铅HK-2细胞中XIAP的表达明显低于对照组,且随着铅浓度的增高而降低,碘化钾能够抑制XIAP的表达的降低;染铅HK-2细胞的凋亡率明显高于对照组,且随着铅浓度的增高而升高(p<0.05/p<0.01),碘化钾能够降低染铅细胞的凋亡率(p<0.05/p<0.01)。结论:碘化钾能够上调染铅HK-2细胞中抗凋亡蛋白XIAP的表达,降低凋亡率,在一定程度上降低铅对细胞的损伤。
AIM: To investigate the effect of potassium iodide on XIAP expression and apoptosis in lead-induced renal tubular epithelial cells (HK-2), and to explore its possible mechanism to resist lead-induced neo-tubule epithelial cell injury. Methods: The expression of XIAP in HK-2 cells and control cells was detected by using cell histochemistry. The apoptosis was detected by flow cytometry. Results: The expression of XIAP in lead-exposed HK-2 cells was significantly lower than that in control group, and decreased with the increase of lead concentration. Potassium iodide could inhibit the decrease of XIAP expression. The apoptosis rate of lead-exposed HK-2 cells was significantly higher than that of control group (P <0.05 / p <0.01), potassium iodide could reduce the apoptosis rate of lead-exposed cells (p <0.05 / p <0.01). Conclusion: Potassium iodide can up-regulate the expression of antiapoptotic protein XIAP in lead-induced HK-2 cells and reduce the apoptosis rate, to a certain extent, reduce the lead injury to cells.
