目的初步探讨T-bet和Eomes的表达与结核病发生的关系。方法分析比较结核患者(TB)与健康对照(HC)、结核菌潜伏感染者(LTBI)以及细菌性肺炎患者(PN)外周血PBMC中T-bet和Eomes的m RNA表达水平,并观察结核患者在抗结核治疗前及抗结核治疗3、6、9和12个月后,外周血PBMC中T-bet和Eomes的m RNA表达水平的变化。结果结核患者(TB)外周血PBMC中T-bet和Eomes的m RNA表达水平显著低于健康对照(HC)、结核菌潜伏感染者(LTBI)以及细菌性肺炎患者(PN)(P<0.05)。同时,在抗结核治疗12个月后,T-bet的表达水平较治疗前显著升高(P<0.05),Eomes的表达水平较治疗前升高,但差异无统计学意义(P>0.05)。结论 T-bet和Eomes两个基因表达水平的下降可能与结核病的发生有关。
Objective To investigate the relationship between the expression of T-bet and Eomes and the occurrence of tuberculosis. Methods The mRNA expression of T-bet and Eomes in peripheral blood of tuberculosis patients (TB) and healthy controls (HC), tuberculosis latent infection (LTBI) and bacterial pneumonia patients (PN) were analyzed and compared. Changes in T-bet and Eomes m RNA expression levels in peripheral blood PBMCs 3, 6, 9 and 12 months after anti-TB treatment and anti-TB treatment. Results The mRNA expression levels of T-bet and Eomes in peripheral blood of patients with tuberculosis (TB) were significantly lower than those of healthy controls (HC), LTBI patients and patients with bacterial pneumonia (P <0.05) . At the same time, the expression of T-bet increased significantly (P <0.05) and the expression of Eomes increased 12 months after anti-TB therapy compared with before treatment, but the difference was not statistically significant (P> 0.05) . Conclusion The decrease of T-bet and Eomes gene expression may be related to the occurrence of tuberculosis.
