目的测定当前鄱阳湖区日本血吸虫毛蚴对吡喹酮的敏感性,提供敏感性监测的基线数据。方法将毛蚴分别暴露于5×10-5、10-5、5×10-6、10-6、5×10-7、10-7 mol/L的吡喹酮溶液中,作用0、1、3、5 min后滴加卢戈氏碘液,待自然静置40 min后,观察毛蚴的运动及形态学变化。结果毛蚴暴露于5×10-5、10-5 mol/L的吡喹酮溶液中,作用0min时均100%变形;暴露于10-7 mol/L的吡喹酮溶液中,作用5 min变形率为(3.5±0.6)%。暴露于5×10-6 mol/L的吡喹酮溶液中作用0、1、3 min后毛蚴变形率为(59.7±7.8)%、(84.4±6.6)%、100%;暴露于10-6 mol/L的吡喹酮溶液中作用0、1、3、5 min后毛蚴变形率为(27.7±4.1)%、(56.2±4.8)%、(84.5±2.5)%、(97.4±1.8)%;暴露于5×10-7mol/L的吡喹酮溶液中作用0、1、3、5 min后,变形率为0%、(5.1±2.1)%、(27.1±2.4)%、(47.5±3.7)%。暴露于10-6mol/L的吡喹酮溶液中作用0、1、3、5 min后各疫区组间的毛蚴变形率差异无统计学意义(F=1.421,P=0.253),不同时间下毛蚴变形率差异有统计学意义(P<0.001)。结论鄱阳湖区血吸虫病重疫区日本血吸虫毛蚴对吡喹酮的敏感性无明显差异。将毛蚴移入10-6 mol/L的吡喹酮溶液中3 min或5 min,镜下观察其变形率,可用于现场监测鄱阳湖区日本血吸虫对吡喹酮的敏感性。
Objective To determine the susceptibility of Schistosoma japonicum mirabilis to praziquantel in the current Poyang Lake region and to provide baseline data for sensitivity monitoring. Methods The miracidia were exposed to praziquantel solution of 5 × 10-5, 10-5, 5 × 10-6, 10-6, 5 × 10-7 and 10-7 mol / L, respectively, After 3,5 min, Lugol’s iodine solution was dropped. After 40 min of natural standing, the movement and morphological changes of miracidia were observed. Results Mirabilis was exposed to 5 × 10-5 and 10-5 mol / L of praziquantel for 100 min at 0 min. After exposed to 10-7 mol / L praziquantel for 5 min, The rate was (3.5 ± 0.6)%. After being exposed to 5 × 10-6 mol / L praziquantel solution for 0, 1 and 3 min, the rate of miosis was (59.7 ± 7.8)%, (84.4 ± 6.6)% and 100% (27.7 ± 4.1)%, (56.2 ± 4.8)%, (84.5 ± 2.5)%, (97.4 ± 1.8)%, respectively in praziquantel solution treated with 0.1, 1, (5.1 ± 2.1)%, (27.1 ± 2.4)% and (47.5 ±)% respectively after being exposed to 5 × 10-7mol / L praziquantel for 0,1,3 and 5 min. 3.7)%. Conclusion There is no significant difference in the sensitivity of Schistosoma japonicum mirabilis to praziquantel in severe flooding area of Poyang Lake. The miracidia was transferred into 10-6 mol / L praziquantel solution for 3 min or 5 min, and its deformation rate was observed microscopically. It could be used to monitor the susceptibility of S. japonicum to praziquantel in the Poyang Lake area.
