目的 :采用UPLC法测定11批不同产地五指毛桃中补骨脂素的含量,为其质量标准的建立提供一定参考依据。方法 :采用XBridge TM BEH C_(18)色谱柱(100 mm×4.6 mm,2.5μm),流动相:乙腈—水(40∶60),流速:0.5 m L/min,柱温:35℃,检测波长:245 nm。结果:补骨脂素对照品在0.78~78μg之间线性范围良好,线性回归方程Y=54.227X+90.378(r=0.9999),平均加样回收率为99.6%,RSD=2.05%(n=6)。各产地的五指毛桃含量在0.039%~0.070%之间,其中,含量排名前三的产地为:广东河源、广东从化、广东翁源,分别为0.070%、0.063%、0.058%。结论:不同产地五指毛桃的补骨脂素含量差异较大。UPLC测定方法出峰时间快,操作简单、准确,可为五指毛桃质量标准建立提供方法学支持。
OBJECTIVE: To determine the content of psoralen in 11 batches of peaches from different habitats by UPLC, and to provide a reference for the establishment of its quality standard. METHODS: The mobile phase was acetonitrile-water (40:60), the flow rate was 0.5 m L / min, the column temperature was 35 ℃, the detection was performed on a XBridge BEH C 18 column (100 mm × 4.6 mm, 2.5 μm) Wavelength: 245 nm. Results: The standard range of psoralen was 0.78 ~ 78μg, the linear regression equation was Y = 54.227X + 90.378 (r = 0.9999), the average recovery was 99.6%, RSD was 2.05% (n = 6 ). The five-finger peach content in all the producing areas ranged from 0.039% to 0.070%. Among them, the top three originated areas were Guangdong Heyuan, Guangdong Conghua and Guangdong’s Wengyuan, with 0.070%, 0.063% and 0.058% respectively. Conclusion: The content of psoralen in fingernails from different areas is quite different. UPLC determination of the peak time is fast, simple, accurate, peach quality standards for the establishment of providing methodological support.
