采用正交试验和浸渍法研究了酶浓度、底物浓度、反应时间等5个因素对中黑盲蝽3龄若虫乙酰胆碱酯酶(AChE)活性的影响和7种农药对中黑盲蝽3龄若虫的毒力,以探讨中黑盲蝽Adel-phocoris suturalis Jakovlev AChE活性测定的最佳反应条件及其对不同药剂的敏感度。测定中黑盲蝽若虫AChE活性的最适反应条件:酶浓度为8头/mL、底物浓度为4mmol/L、反应时间为5min、反应温度为40℃,pH值为6.5。毒死蜱、辛硫磷、乙酰甲胺磷、灭多威、敌敌畏、马拉硫磷、丁硫克百威对中黑盲蝽3龄若虫的LC50值分别为20.50、22.97、32.20、46.63、61.52、73.47、153.41μg/mL。7种药剂对中黑盲蝽若虫体内AChE的抑制能力由大到小依次为毒死蜱、辛硫磷、乙酰甲胺磷>灭多威、敌敌畏、马拉硫磷>丁硫克百威。研究表明,毒死蜱、辛硫磷、乙酰甲胺磷是防治中黑盲蝽较有潜力的药剂。
The effects of enzyme concentration, substrate concentration and reaction time on acetylcholinesterase (AChE) activity of Nystagmus nymphs (3rd instar nymphs) and the effect of 7 pesticides on the 3rd instar larvae Nymphs to investigate the optimal reaction conditions and the sensitivity to different agents in the determination of AChE activity in Adel-phocoris suturalis Jakovlev. The optimum reaction conditions for the determination of AChE activity in Nymphs nymphs were: enzyme concentration of 8 heads / mL, substrate concentration of 4mmol / L, reaction time of 5min, reaction temperature of 40 ℃ and pH value of 6.5. The LC50 values of chlorpyrifos, phoxim, acephate, methomyl, dichlorvos, malathion and metsulfuron-methyl were 3.50, 73.47, 153.41 μg / mL. The inhibition ability of AChE from 7 kinds of agents to the nymphs of Lagers nymphs was chlorpyrifos, phoxim, acephate> methomyl, dichlorvos, malathion> carbosulfan. Studies have shown that chlorpyrifos, phoxim and acephate are potential agents for the control of Apine melaleuca.
