目的 :从整体水平探讨瑞香狼毒水提物 (SCLA)的抗癌机理。方法 :以不同剂量SCLA灌胃给药后 ,在不同时间采集小鼠血清 ,处理体外培养的人肝癌BEL 740 2细胞 ,用MTT比色法和集落形成试验观察细胞增殖能力。结果 :SCLA5、10、2 0g·kg-1给药 1、2、3h后的药物血清处理肿瘤细胞后 ,其MTT转化率和克隆形成率均显著降低 ;给药 2h后的药物血清表现出更强的抑制肿瘤细胞增殖作用。结论 :直接抑制癌细胞增殖是瑞香狼毒的主要抗癌机理
Objective : To explore the anti-cancer mechanism of Stellera chamaejasme aqueous extract (SCLA) from the overall level. METHODS: Following intragastric administration of different doses of SCLA, mouse sera were collected at different times to treat human hepatocellular carcinoma BEL 740 2 cells in vitro. Cell proliferation was assessed by MTT assay and colony formation assay. RESULTS: MTT conversion and colony formation rate were significantly decreased after treatment with SCLA 5, 10, 20 g·kg-1 drug serum for 1, 2, and 3 h after treatment. The drug serum after 2 h administration showed more Strong inhibition of tumor cell proliferation. Conclusion : Direct inhibition of cancer cell proliferation is the main anticancer mechanism of Stellera chamaejasme
