目的建立颠茄酊的HPLC特征图谱及东莨菪内酯的含量测定方法。方法采用Welch XB-C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.05%磷酸,梯度洗脱,流速:1.0 m L·min?1;柱温:30℃;检测波长:344 nm。结果建立了颠茄酊HPLC特征图谱,确定了6个共有峰,指认了其中3个共有峰的成分分别为山柰素-3-O-半乳糖-(6→1)鼠李糖-7-O-葡萄糖苷、东莨菪内酯和芦丁。东莨菪内酯进样量在11.18~670.5 ng(r=1.000 0)内线性关系良好,平均加样回收率为104.2%。结论特征图谱结合东莨菪内酯的定量测定,能更好地控制颠茄酊的质量。
OBJECTIVE To establish a HPLC method for the determination of belladonna tincture and the method for the determination of scopoletin. The method was performed on a Welch XB-C18 (250 mm × 4.6 mm, 5 μm) column with a mobile phase of methanol-0.05% phosphoric acid with gradient elution at a flow rate of 1.0 mL · min -1 and a column temperature of 30 ° C. Detection wavelength: 344 nm. Results HPLC profiles of belladonna tincture were established and six common peaks were identified. Three of the common peaks were identified as benthon-3-O-galactose- (6 → 1) rhamnose-7- O-glucoside, scopoletin and rutin. The injection of scopoletin showed good linearity in the range of 11.18-670.5 ng (r = 1.000 0) with an average recovery rate of 104.2%. Conclusion The characteristic map combined with the quantitative determination of scopoletin can better control the quality of belladonna tincture.
