目的:研究人类eag相关基因(herg基因)及其表达的hERG蛋白在喉癌中的表达情况。方法:应用免疫组织化学方法和real-time PCR技术对30例喉癌组织标本及20例癌旁2cm黏膜组织中的herg基因及hERG蛋白的表达进行检测。结果:1免疫组织化学分析hERG蛋白在喉癌组织的阳性率为76.7%(23/30),而在癌旁2cm黏膜组织中的阳性率为10.0%(2/20),两者比较差异有统计学意义(P<0.05)。2real-time PCR分析喉癌组织中herg基因的mRNA表达是癌旁2cm黏膜组织的3.25倍。结论:herg基因在喉癌中呈高水平表达可能与肿瘤的发生、发展有关。
Objective: To study the expression of human eag related gene (herg gene) and its expressed hERG protein in laryngeal carcinoma. Methods: The expression of herg gene and hERG protein in 30 specimens of laryngeal carcinoma and 20 specimens of 2 cm adjacent mucosa were detected by immunohistochemistry and real-time PCR. The positive rate of hERG protein in laryngeal carcinoma was 76.7% (23/30) by immunohistochemical staining and 10.0% (2/20) in 2 cm mucosa adjacent to carcinoma, the differences were statistically significant Statistical significance (P <0.05). 2real-time PCR analysis of laryngeal cancer herg gene mRNA expression was 3.25 times the adjacent 2cm mucosal tissue. Conclusion: The high level of herg gene expression in laryngeal carcinoma may be related to the occurrence and development of tumors.
