目的:研究狗舌草提取物对U266细胞的细胞凋亡作用及其机制。方法:U266细胞株与狗舌草提取物共同培养,电镜观察细胞凋亡形态,DNA琼脂糖凝胶电泳观察凋亡条带。用流式细胞仪测定细胞周期及凋亡率。结果:狗舌草提取物作用U266细胞,电镜观察到细胞凋亡,DNA琼脂糖凝胶电泳出现凋亡条带,狗舌草提取物影响U266细胞周期,G0/G1期细胞减少,S期、G2/M期细胞增加,凋亡细胞增加,用Annexin V/PI染色流式细胞仪检测,均显示细胞凋亡与狗舌草提取物有剂量依赖关系。结论:体外实验证实,狗舌草提取物对266细胞有细胞凋亡作用,存在剂量依赖关系。
Objective: To study the apoptosis and its mechanism of Rugosa extract on U266 cells. Methods: U266 cells were cultured with Ragwort extract, and the morphological changes of cells were observed by electron microscope. The apoptotic bands were observed by DNA agarose gel electrophoresis. Cell cycle and apoptosis rate were determined by flow cytometry. Results: Apoptosis of U266 cells was observed by Rugosa extract and apoptotic bands were observed by electron microscopy. Apoptotic bands were observed by DNA agarose gel electrophoresis. Rugosae extract affected cell cycle of U266 cells, decreased cells in G0 / G1 phase, S phase, G2 / M phase cells increased, apoptotic cells increased, with Annexin V / PI staining flow cytometry showed apoptosis and Radix leptin extract in a dose-dependent manner. Conclusion: In vitro experiments confirmed that Ragwort extract on 266 cells apoptosis, in a dose-dependent manner.
