目的观察肾上腺素(EPI)对内毒素(LPS)血症大鼠体内肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-10(IL-10)的影响。了解其在大鼠体内是否有抗炎作用。方法2006-11—2007-09,于广州医学院实验动物中心将50只SD大鼠随机分为5组(每组10只):正常对照组大鼠静脉输注生理盐水2·4mL·kg-1·h-1;LPS组大鼠静脉注射内毒素6mg·kg-1后静脉输注生理盐水2·4mL·kg-1·h-1;低、中和高剂量EPI组大鼠静脉注射内毒素6mg·kg-1后,分别静脉输注肾上腺素0·12μg·kg-1·min-1、0·3μg·kg-1·min-1和0·6μg·kg-1·min-1。在当即、第2小时、第6小时三个时点取血,检测血清TNF-α、IL-1β和IL-10水平。结果与对照组比较,LPS组大鼠血清TNF-α、IL-1β和IL-10水平均升高。与LPS组比较,高剂量EPI组血清TNF-α水平明显下降(P<0·05),血清IL-10水平明显升高(P<0·05),而血清IL-1β水平在两组间差异无显著性(P>0·05)。TNF-α、IL-1β和IL-10水平在中低剂量EPI组与LPS组间差异无显著性(P>0·05)。结论肾上腺素对内毒素血症大鼠具有抗炎作用,提示肾上腺素对于调控脓毒症时过度炎症反应具有潜在的临床应用价值。
Objective To observe the effects of epinephrine (EPI) on tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-10 (IL-10) in rats with endotoxin (LPS) Impact. METHODS: From January 2006 to July 2007, 50 SD rats were randomly divided into 5 groups (10 in each group) in Experimental Animal Center of Guangzhou Medical College: The rats in normal control group received intravenous infusion of normal saline (2.4 mL · kg- 1 · h-1. The rats in LPS group were intravenously injected with 4 mg · kg-1 of endotoxin and 2 · 4 mL · kg-1 · h-1 of saline. The rats in low, middle and high dose EPI group were injected intravenously After intravenous injection of 6 mg · kg-1 of toxin, epinephrine (0.12 μg · kg-1 · min-1, · 3 μg · kg-1 · min-1 and 0.6 μg · kg-1 · min- Blood samples were taken immediately after the first hour, the second hour and the sixth hour, and the levels of serum TNF-α, IL-1β and IL-10 were measured. Results Compared with the control group, the levels of serum TNF-α, IL-1β and IL-10 in LPS group increased. Compared with LPS group, serum TNF-αlevel of high-dose EPI group was significantly decreased (P <0.05), serum IL-10 level was significantly increased (P <0.05), while serum IL-1beta level was lower There was no significant difference (P> 0.05). The levels of TNF-α, IL-1β and IL-10 were not significantly different between low-dose EPI group and LPS group (P> 0.05). Conclusion Epinephrine has an anti-inflammatory effect on endotoxemic rats, suggesting that epinephrine has potential clinical value in the regulation of excessive inflammatory reaction in sepsis.
