目的探讨宫宁颗粒诱导IUD避孕大鼠血管平滑肌细胞(VSMC)凋亡的影响。方法将50只已生育SD雌性大鼠随机分为宫宁颗粒组、消炎痛组、模型组、假手术组、空白对照组,应用光镜观察大鼠子宫组织病理学变化;免疫组化法检测大鼠子宫VSMC中Bcl-2、Bax、Fas和FasL蛋白表达。结果置入IUD后大鼠子宫内膜及血管结构功能发生异常,与模型组比较,宫宁颗粒能显著降低子宫VSMC中Bcl-2蛋白表达,增加Bax、Fas、FasL蛋白表达,并降低Bcl-2/Bax值(P<0.01),且效果明显优于消炎痛组(P<0.01)。结论宫宁颗粒可减轻大鼠子宫内膜形态学损伤,消除子宫血管构筑的异常,其机制可能是通过升高Bax、Fas、FasL蛋白表达,降低Bcl-2蛋白表达,诱导子宫VSMC凋亡,进而达到防治置环后异常子宫出血的目的。
Objective To investigate the effects of Gongning granule on the apoptosis of vascular smooth muscle cells (VSMCs) of IUD contraceptive rats. Methods Fifty SD female SD rats were randomly divided into three groups: Gongning granule group, indomethacin group, model group, sham operation group and blank control group. The pathological changes of uterus were observed with light microscope. Expression of Bcl-2, Bax, Fas and FasL in rat uterine VSMCs. Results Compared with model group, Gongning granule could significantly reduce the expression of Bcl-2 protein, increase the expression of Bax, Fas and FasL in uterine VSMC and decrease the expression of Bcl- 2 / Bax (P <0.01), and the effect was better than indomethacin group (P <0.01). Conclusion Gongning granule can reduce the morphological damage of endometrium in rats and eliminate the abnormality of uterine vascular architecture. The mechanism may be through increasing the expression of Bax, Fas and FasL, decreasing the expression of Bcl-2 and inducing the apoptosis of uterine VSMC, So as to achieve the purpose of preventing and treating abnormal uterine bleeding after setting ring.
