目的:探讨新疆阿魏乙酸乙酯部位对人结肠癌Caco-2细胞周期和凋亡的影响,并对抑癌基因PTEN、p53进行考察.方法:将新疆阿魏乙酸乙酯部位用完全培养液配成高、中、低浓度的溶液(15、30、60 μg/mL),分别作用于Caco-2细胞,采用流式细胞仪对各组细胞所处周期和凋亡情况进行检测,采用RT-PCR、Western Blot方法检测调控细胞周期、凋亡的PTEN、p53基因和蛋白的表达水平.结果:新疆阿魏乙酸乙酯各剂量组使Caco-2细胞大量阻滞在G0/G1期(P<0.05),并且各给药组Caco-2细胞的凋亡率均明显高于空白组(P<0.01);RT-PCR、Western B10t检测结果显示各给药组Caco-2细胞的PTEN、p53的基因表达水平均显著增高(P<0.01),PTEN、p53的蛋白相对表达水平显著增高(P<0.05或P<0.01),与顺铂对Caco-2细胞的作用效果相似.结论:新疆阿魏乙酸乙酯部位可以使人结肠癌Caco-2细胞分裂阻滞在G0/G1期,促进Caco-2细胞凋亡,并且显著提高抑癌基因PTEN、p53的表达.“,”Objective:To study the effect of Ferula sinkiangensis K.M.Shen optimization ethyl acetate parts on colon Caco-2 and the influence of cell cycle and apoptosis of tunor suppressor gene PTEN,p53.Methods:The Caco-2 cells were intervened by Ferula sinkiangensis K.M.Shen ethyl acetate parts with complete nutrient solution with the concentration of 15,30,60 μg/nL solution respectively.Apoptosis for each cell cycle were detected by flow eytometry instrument.Apoptosis and cell cycle related tumor suppressor gene PTEN,the relative expression level of p53 gene and protein were detected by rt-pcr and Western Bloting detection.Results:A large number of Caco-2 cells were in G0/G1 phase in Ferula sinkiangensis K.M.Shen ethyl acetate group (P<0.05),and medicine group showed higher Caco-2 cells apoptosis than the blank group (P<0.01);Rt-pcr and Western Bloting testing results showed that Caco-2 cells PTEN and p53 gene expression levels were significantly increased in the medicine group (P<0.01),and that the relative expression level of PTEN and p53 protein is also significantly increased (P<0.05 or P<0.01);the effects were similar with the positive drug cisplatin.Conclusion:Ferula sinkiangensis K.M.Shen ethyl acetate parts can block the Caco-2 cell division of human colon cancer in G0/G1 phase,promote Caco-2 cell apoptosis,and increase the expression of tumor suppressor gene PTEN and p53 gene has important link..
