目的:研究1700001O22RIK(RIKEN c DNA 1700001O22)基因在小鼠睾丸的表达特征,结合生物信息学分析初步探讨其功能。方法:通过分析小鼠基因表达谱芯片,发现1700001O22RIK基因特异性表达于睾丸组织。采用RT-PCR、实时PCR、Western印迹及免疫组化等方法分析该基因在成年C57BL/6J小鼠各组织中的表达特征,以及不同发育阶段小鼠的睾丸组织中的表达变化。利用生物信息学软件对该基因进行相关生物信息学分析。结果:1700001O22RIK基因在小鼠睾丸组织中呈现特异性表达,表达水平具有年龄依赖性,在成年期表达最强;1700001O22RIK蛋白在成年小鼠睾丸组织中主要定位于精原细胞、精母细胞及圆形精子细胞。经生物信息学分析,1700001O22RIK基因编码蛋白氨基酸序列在人和小鼠具有高度同源性,提示该基因在哺乳动物进化过程中十分保守。结论:1700001O22RIK属于睾丸特异性基因,主要表达于生精小管的精原细胞、精母细胞及圆形精子细胞,提示其可能参与精子发生的调节。
Objective: To study the expression of 1700001O22RIK (RIKEN c DNA 1700001O22) gene in mouse testis and to explore the function of RIKEN c DNA 1700001O22 gene in combination with bioinformatics analysis. Methods: By analyzing mouse gene expression profiling chips, it was found that 1700001O22RIK gene was specifically expressed in testicular tissue. RT-PCR, real-time PCR, Western blot and immunohistochemistry were used to analyze the expression characteristics of the gene in different tissues of adult C57BL / 6J mice and the changes of testis in different developmental stages. The bioinformatics software was used to analyze the gene. Results: The 1700001 O22RIK gene was specifically expressed in mouse testis tissue and the expression level was age-dependent, which was the strongest in adult mice. The 1700001 O22RIK protein mainly localized in spermatogonia, spermatocytes and round Sperm cells. Bioinformatics analysis, 1700001O22RIK gene encoded protein amino acid sequence in human and mouse has a high degree of homology, suggesting that the gene in the mammalian evolutionary process is very conservative. CONCLUSION: 1700001 O22RIK belongs to the testis-specific gene and is mainly expressed in spermatogonia, spermatogonia and round spermatids, suggesting that it may be involved in the regulation of spermatogenesis.
