目的探讨溶血和乳糜微粒对ALT检测的影响。方法根据中华人民共和国卫生行业标准(WS/T)和美国临床和实验室标准研究院(CLSI)相关文件,进行干扰实验验证。结果干扰实验结果显示,样本在反复冻融1次以上和震荡2 min以上时,以及外源性干扰物血红蛋白(HB)>2.5 g/L时ALT检测均产生正干扰,乳糜微粒>5 g/L对ALT检测有明显负干扰。结论血红蛋白产生的是正干扰,乳糜微粒产生的是负干扰,通过评估溶血和脂血标本的干扰方向和干扰偏差的大小,对如何处理生物参考区间上限附近的标本具有指导作用。
Objective To investigate the effect of hemolysis and chylomicrons on the detection of ALT. Methods According to the health industry standard of the People’s Republic of China (WS / T) and the relevant documents of the American Clinical and Laboratory Standards Institute (CLSI), interference experiments were conducted. Results The results of interference experiments showed that there was positive interference in the ALT assay when cholera particles> 5 g / L were exposed to repeated freezing and thawing for more than 1 second and shaking for more than 2 minutes, and exogenous interferon hemoglobin (HB)> 2.5 g / L has a clear negative interference with ALT detection. Conclusion Hemoglobin produces positive interference and chylomicrons produce negative interference. By assessing the direction of interference and the size of interference deviation in hemolysis and lipids, it is instructive for the treatment of specimens near the upper limit of biological reference interval.
