目的 探讨体外光动力学治疗 (PDT)引起鼻咽癌 (NPC)细胞死亡的作用机制。方法 对鼻咽癌细胞株HNE1体外光动力学治疗前后形态学变化进行动态观察。结果 对照组HNE1细胞培养 3h ,6h ,12h ,2 4h和 4 8h后细胞形态与正常培养组HNE1细胞形态相比较无明显改变。PDT 12h后 ,实验组HNE1细胞开始变形皱缩 ,细胞界限模糊 ,细胞透光度和立体感较差 ,治疗 2 4~ 4 8h后细胞发生凋亡。结论 体外光动力学治疗对HNE1具有明显的杀伤效应并诱导HNE1细胞凋亡。PDT治疗鼻咽癌可能通过其诱导NPC细胞凋亡而发生效应
Objective To investigate the mechanism of photodynamic therapy (PDT) induced cell death in nasopharyngeal carcinoma (NPC). Methods Morphological changes of nasopharyngeal carcinoma cell line HNE1 before and after photodynamic therapy were dynamically observed. Results The morphology of HNE1 cells in the control group at 3 h, 6 h, 12 h, 24 h, and 48 h did not change significantly compared with the normal culture group. After 12 h of PDT, HNE1 cells in the experimental group began to deform and shrink, cell boundaries were blurred, cell transmittance and stereoscopic sensation were poor, and apoptosis occurred in cells after 24 to 48 hours of treatment. Conclusion In vitro photodynamic therapy has a significant killing effect on HNE1 and induces apoptosis in HNE1 cells. PDT treatment of nasopharyngeal carcinoma may induce apoptosis through NPC cells
