目的比较前突触终末和树突棘之间突触连接的能动性。方法运用GFP转染小脑Purkinje细胞、DiI标记平行纤维双标记技术标记样本,双光子显微镜进行活体观察。结果前突触终末与树突棘之间有4种不同的接触方式;树突棘的能动性远大于前突触终末;树突棘和树突棘表面的伪足可主动搜寻附近的前突触终末,与之形成突触连结。结论树突棘的能动性远大于前突触终末,树突棘及其表面的伪足有高度的能动性,提示他们在突触发生过程中发挥特殊作用。
Objective To compare the motility of synaptic connections between presynaptic terminals and dendritic spines. Methods Purkinje cells were transfected with GFP. DiI-labeled parallel fiber double labeling technique was used to label the samples. Two-photon microscopy was used to observe the morphology of Purkinje cells. Results There were 4 different contact modes between the presynaptic terminal and the dendritic spine. The densities of dendritic spines were much larger than those of the presynaptic terminals. The pseudopodia on the surface of dendritic spines and dendritic spines could actively search nearby Synaptic terminal, with the formation of synaptic connections. Conclusion The dendritic spine is much more motile than the presynaptic terminal. The dendritic spines and their pseudopods on the surface have a high degree of motility, suggesting that they play a special role in synaptogenesis.
