目的:探讨HPD-100大孔吸附树脂对(-)-表儿茶素纯化的最佳工艺。方法:通过动态吸附-解吸相结合的方法,以(-)-表儿茶素吸附量、解吸率为评价指标,采用高效液相色谱法进行含量测定,综合评定最佳工艺。结果:HPD-100大孔吸附树脂纯化效果较好,其最佳工艺的条件为:上样液为180mL,吸附速率为1mL.m in-1,水洗量为1倍的树脂湿体积,洗脱液为30%乙醇180mL,解吸速率为1mL.m in-1。肺舒通提取液纯化后,(-)-表儿茶素的质量分数为纯化前的5.07倍。结论:HPD-100大孔吸附树脂纯化(-)-表儿茶素的最佳工艺稳定、较好。
Objective: To investigate the best process for the purification of (-)-epicatechin by HPD-100 macroporous resin. Methods: The method of dynamic adsorption-desorption combined with (-)-epicatechin adsorption and desorption was used as evaluation index. The content was determined by high performance liquid chromatography and the best process was evaluated comprehensively. Results: The purification effect of HPD-100 macroporous adsorption resin was better. The optimal conditions for the process were as follows: 180 mL of sample solution, adsorption rate of 1 mL.m in-1, and the water volume of the resin was doubled. The liquid was 180 mL of 30% ethanol and the desorption rate was 1 mL.m in-1. After purification of Feishutong extract, the mass fraction of (-)-epicatechin was 5.07 times before purification. Conclusion: The best process for the purification of (-)-epicatechin from HPD-100 macroporous adsorption resin is stable and good.
