目的研究沙格雷酯对野百合碱模型大鼠肺动脉高压的改善作用及对肺动脉活化T细胞核因子3(NFAT3)表达的影响。方法实验分为对照组、野百合碱诱导的肺动脉高压模型组、沙格雷酯干预组,每组6只大鼠。建立野百合碱诱导的肺动脉高压大鼠模型,沙格雷酯干预组给予5-HT2A受体拮抗剂沙格雷酯干预(100 mg/kg)4周。分别测定肺动脉压力,计算右心指数,肺动脉组织HE染色评价肺动脉壁增厚程度,Western blot检测肺动脉组织NFAT3蛋白表达水平。结果与肺动脉高压模型组相比,沙格雷酯干预组的平均肺动脉压力和右心指数降低,肺动脉壁增厚程度减轻,肺动脉NFAT3蛋白表达水平下调(P<0.05)。结论沙格雷酯可改善野百合碱动物模型的肺动脉高压程度,并下调肺动脉高压肺动脉的NFAT3蛋白表达水平。
Objective To study the effect of sarpogrelate on pulmonary hypertension induced by monocrotaline and its effect on the expression of nuclear factor-3 (NFAT3) in pulmonary artery activated T cells. Methods The experiment was divided into control group, monocrotaline-induced pulmonary hypertension model group and shagrelone intervention group, 6 rats in each group. The rat model of pulmonary hypertension induced by monocrotaline was established. The shalegreotide intervention group was given sarprelide (100 mg / kg) as a 5-HT2A receptor antagonist for 4 weeks. The pulmonary artery pressure was measured, the right heart index was calculated, the pulmonary artery wall thickening was evaluated by HE staining, and the expression of NFAT3 protein in pulmonary artery was detected by Western blot. Results Compared with pulmonary arterial hypertension group, the mean pulmonary artery pressure and right heart index in the sharerethane intervention group were decreased, the thickening of the pulmonary artery wall was alleviated, and the expression of NFAT3 protein in the pulmonary artery was decreased (P <0.05). Conclusion Sagulide can improve the pulmonary hypertension and reduce the expression of NFAT3 in pulmonary artery of pulmonary hypertensive rats.
