目的:探讨枳实消痞丸方对大鼠胃及十二指肠黏膜胆囊收缩素及其A受体基因表达的影响.方法:♂Wistar大鼠20只,分为对照及用药组,每组10只.分别以1.5mL生理盐水及枳实消痞丸方水煎制剂灌胃4wk.处死后取材,用RT-PCR法检测十二指肠黏膜CCKmRNA及胃底、胃窦肌肉组织CCK-A受体mRNA表达水平,并以同管所扩β-actin作为内参照进行半定量比较.结果:中药组大鼠十二指肠黏膜CCKmRNA及胃底CCK-A受体mRNA的表达水平明显较对照组降低(1.13±0.26vs0.67±0.10,P<0.001;7.30±2.61vs3.37±1.07,P<0.01),但胃窦部的CCK-A受体mRNA的表达水平明显增高(1.22±0.24vs3.25±1.11,P<0.001).结论:枳实消痞丸方可能通过对CCKmRNA及CCK-A受体mRNA表达水平的影响,从而起到对消化道动力的某些影响及临床上的一些治疗作用.
Objective: To investigate the effect of Gushi Xiaotanwan Prescription on the expression of cholecystokinin and its receptor A gene in rat gastric and duodenal mucosa. Methods: Twenty Wistar rats were divided into control and drug groups, each group Ten rats were given intragastric administration of 1.5 mL normal saline and Zhishi Xiaotan Wanfang decoction respectively for 4 weeks. Samples were taken after sacrifice and RT-PCR was used to detect CCK mRNA in the duodenal mucosa and CCK-A in the stomach and antrum muscle tissue. Receptor mRNA expression level was compared with β-actin expanded as an internal reference. Results: The expression of CCK mRNA and CCK-A receptor mRNA in the gastric mucosa of the duodenal mucosa of the TCM group was significantly lower than that of the control group. The group was decreased (1.13±0.26 vs 0.67±0.10, P<0.001; 7.30±2.61 vs 3.37±1.07, P<0.01), but the expression level of CCK-A receptor mRNA in gastric antrum was significantly higher (1.22±0.24). (vs3.25±1.11, P<0.001). Conclusion: The effect of Gushi Xiaolan Wanfang on the expression of CCK mRNA and CCK-A receptor mRNA may play a role in the gastrointestinal motility and clinically Some treatment effects.
