目的研究硫化氢(hydrogen sulfide,H2S)缺血后处理能否减轻家兔心肌缺血/再灌注损伤。方法将24只家免随机分为缺血/再灌注(IP)组、缺血后处理(IPO)组和硫氢化钠(H2S供体)后处理(NPO)三组,每组8只。观察各组家兔的心率(HR)、左室发展压(LVDP)、丙二醛(MDA)、超氧化物歧化酶(SOD)和心肌梗死面积变化。结果缺血180min后,NPO组心率较IP组、IPO组明显下降(P分别<0.05);与IP组相比,IPO组及NPO组LVDP显著升高(P分别<0.05,<0.01),且NPO组LVDP升高较IPO组明显,差异有统计学意义(P<0.05)。②与IP组比较,IPO组及NPO组血清中MDA含量显著下降(P分别<0.05,<0.01),同时SOD活性显著上升,差异有统计学意义(P分别<0.05,<0.01);NPO组MDA含量较IPO组降低,差异有统计学意义(P<0.05),而SOD活性增高,差异有统计学意义(P<0.05)。③与IP组心肌坏死面积(45.2±4.3)%比较,IPO(34.3±6.2)%和NPO(23.4±4.6)%显著减少再灌注家免心肌坏死面积(P分别<0.05),且NPO组比IPO组心肌坏死面积减小更明显,差异有统计学意义(P<0.05)。结论 H2S后处理能减轻家兔心肌缺血/再灌注损伤,对心肌有保护作用。
Objective To investigate whether ischemic postconditioning with hydrogen sulfide (H2S) can reduce myocardial ischemia / reperfusion injury in rabbits. Methods Totally 24 children were randomly divided into three groups: ischemia / reperfusion (IP) group, IPO group and post-treatment of sodium hydrogen sulfide (H2S donor). The changes of heart rate (HR), left ventricular development pressure (LVDP), malondialdehyde (MDA), superoxide dismutase (SOD) and myocardial infarct area in each group were observed. Results Compared with IP group, the LVDP in IPO group and NPO group was significantly higher than that in IP group (P <0.05, <0.01 respectively), and the heart rate in NPO group was significantly lower than that in IP group and IPO group LVDP increased significantly in NPO group compared with IPO group, the difference was statistically significant (P <0.05). ②Compared with IP group, serum MDA levels in IPO group and NPO group were significantly decreased (P <0.05, <0.01 respectively), while SOD activity increased significantly (P <0.05, <0.01 respectively) MDA content was lower than IPO group, the difference was statistically significant (P <0.05), while SOD activity increased, the difference was statistically significant (P <0.05). (3) IPO (34.3 ± 6.2)% and NPO (23.4 ± 4.6)% significantly reduced the area of myocardial necrosis without reperfusion injury (P <0.05, respectively) compared with 45.2 ± 4.3% The area of myocardial necrosis in IPO group decreased more obviously, the difference was statistically significant (P <0.05). Conclusion H2S postconditioning can attenuate myocardial ischemia / reperfusion injury in rabbits and protect myocardium.
