目的研究小鼠在胚胎及生后发育不同阶段肺β-防御素1(mBD-1)的表达。方法孕鼠随机分成2组:对照组和实验组。实验组于胚胎提取及出生前24h腹腔注射LPS,对照组以等剂量生理盐水代替。各组分别于14.5、17.5 dpc(days post coition,交配后天数)、生后0d、生后4d取肺组织进行石蜡包埋、连续切片,并进行mBD-1的免疫组化染色,用图像分析系统进行定量分析,并进行统计学处理。结果对照小鼠从14.5d胚胎至出生后4 d各时期肺组织中均存在mBD-1阳性表达,且呈增加趋势(P<0.05);而同一时间点实验组阳性表达略高于对照组,但与对照组相比差异无统计学意义(P>0.05)。结论本实验表明不同发育阶段小鼠肺组织均表达mBD-1,且其表达呈上升趋势,提示该分子可能是肺组织天然抵抗微生物感染的一个重要分子基础。
Objective To study the expression of lung β-defensin 1 (mBD-1) in different stages of embryo and postnatal development in mice. Methods Pregnant mice were randomly divided into 2 groups: control group and experimental group. The experimental group was intraperitoneally injected with LPS 24 hours before embryo extraction and the control group was replaced by equal dose of saline. The lungs were harvested at 14.5 and 17.5 dpc days after mating, 0 days after birth and 4 days after birth, respectively. The lungs were paraffin-embedded and serially sectioned. The expression of mBD-1 was detected by image analysis System for quantitative analysis, and statistical analysis. Results The expression of mBD-1 in the lung tissue of control mice from 14.5d embryo to 4d after birth was increased (P <0.05), while the positive expression in the experimental group was slightly higher than that in the control group But there was no significant difference compared with the control group (P> 0.05). CONCLUSION: Our results showed that mBD-1 was expressed in the lungs of mice at different developmental stages, and the expression of mBD-1 was up-regulated, suggesting that the molecule may be an important molecular basis of natural resistance to microbial infection in lung tissues.
