目的探讨复方川芎颗粒中药物最佳提取工艺及相关影响因素。方法对复方川芎颗粒采用正交实验找出复方川芎颗粒最佳的提取工艺条件,并采用高效液相色谱法测定复方川芎颗粒中各成分的含量:以150mm×4.6mm作为色谱分析柱,并采用25 75的甲醇-醋酸作为流动相,流动设为10ml/min,检测波长设为280nm作为提取优化条件。结果川芎颗粒最佳的提取工艺为:80%的乙醇以12倍的量采用超声提取2次,每次提取时间为45min。经研究发现芎颗粒在2.02-60.6μg/ml的范围内呈现良好的线性关系,经回归方程分析可知Y=20205X-11452,r=1.00,平均回收率为101.70,RSD=2.78%。结论经改良过的提取工艺可有效提取复方川芎颗粒中的有效成分川芎嗪,提取有效率较高,检测准确快速。在此条件下采用高效液相色谱能有效分离川芎嗪和其他成分。
Objective To investigate the optimal extraction process of compound Chuanxiong Granules and its related factors. Methods The optimum extraction conditions of Compound Chuanxiong Granules were determined by orthogonal experiment on compound Chuanxiong Granules. The content of each component in Chuanxiong Compound Granules was determined by high performance liquid chromatography (HPLC): 150mm × 4.6mm was used as chromatographic column, 25 75 methanol-acetic acid as the mobile phase, flow was set to 10ml / min, detection wavelength was set to 280nm extraction and optimization conditions. Results Chuanxiong particles best extraction process: 80% ethanol 12 times the amount of ultrasound extraction 2 times, each extraction time of 45min. The study found that the Xiong particles in the range of 2.02-60.6μg / ml showed a good linear relationship, the regression equation shows that Y = 20205X-11452, r = 1.00, the average recovery was 101.70, RSD = 2.78%. Conclusion The modified extraction process can effectively extract ligustrazine in Compound Chuanxiong Granules with high extraction efficiency and accurate and rapid detection. In this condition, high performance liquid chromatography can be used to effectively separate ligustrazine and other components.
