目的通过喘可治注射液对细胞因子诱导的杀伤细胞(CIK)的表型及细胞因子分泌情况的影响,探讨喘可治注射液对CIK免疫活性的免疫调节作用。方法分离健康志愿者的外周血单个核细胞,在细胞因子的作用下,体外培养成细胞因子诱导的杀伤细胞(CIK),在培养过程中同时加入喘可治注射液。LDH法检测不同浓度喘可治注射液培养条件下的CIK细胞的抗肿瘤活性。流式细胞术检测CIK细胞表型及细胞内因子的分泌情况变化。Annexin V/PI凋亡试剂盒检测CIK细胞的凋亡情况。结果喘可治注射液能提高CD3+CD56+双阳性的CIK细胞的比例,增强其杀伤肿瘤细胞的活性,促进具有抗肿瘤效应的Th1型的细胞内因子IFN-γ、TNF-α的分泌,而对CD4+CD25+抑制性细胞和Th2型的细胞因子IL-4的分泌影响不大。喘可治注射液对CIK细胞的凋亡没有明显作用。结论喘可治注射液可上调CIK细胞分泌Th1型的细胞因子,增强CIK细胞的抗肿瘤活性。
Objective To investigate the effect of Chuankezhi injection on the immunopotentiation of CIK immunocompetence through the influence of Chuankezi Injection on the phenotype of cytokine-induced killer cells (CIK) and the secretion of cytokines. Methods Peripheral blood mononuclear cells from healthy volunteers were isolated and cultured in vitro into cytokine-induced killer cells (CIKs) under the action of cytokines. Chuankezhi injection was added during the culture process. The antitumor activity of CIK cells cultured with different concentrations of Chuankezhi injection was detected by LDH method. Flow Cytometry to detect CIK cell phenotype and changes in the secretion of intracellular factors. Annexin V / PI apoptosis kit for the detection of CIK cell apoptosis. Results Chuankezhi injection can increase the proportion of CD3 + CD56 + double-positive CIK cells, enhance its activity of killing tumor cells and promote the secretion of Th1-type intracellular factors IFN-γ and TNF-α with anti-tumor effect Had little effect on the secretion of CD4 + CD25 + inhibitory cells and Th2-type cytokine IL-4. Chuankezhi injection had no obvious effect on the apoptosis of CIK cells. Conclusion Chuankezhi injection can up-regulate the secretion of Th1-type cytokines in CIK cells and enhance the anti-tumor activity of CIK cells.
