目的研究金黄色葡萄球菌(简称金葡菌)α溶血素诱导脐静脉内皮细胞的凋亡效应。方法用不同浓度金葡菌α溶血素感染脐静脉内皮细胞,2、4、8和12h后,用流式细胞术检测细胞凋亡率。结果金葡菌α溶血素10、30、90和270ng/ml均可诱导脐静脉内皮细胞发生凋亡,以30ng/ml时诱导细胞凋亡能力最强,作用2h后细胞即发生凋亡,并随着诱导时间的延长,细胞凋亡率逐渐增加。结论金葡菌α溶血素可诱导脐静脉内皮细胞凋亡,并存在明显的时间、剂量效应关系,这可能是金葡菌L型垂直感染影响胎儿宫内发育的重要机理之一。
Objective To study the apoptosis of human umbilical vein endothelial cells induced by Staphylococcus aureus α-hemolysin. Methods Human umbilical vein endothelial cells were infected with different concentrations of S.aureus alpha hemolysin, and the apoptosis rates were detected by flow cytometry after 2, 4, 8 and 12 hours. Results Staphylococcus aureus α hemolysin 10, 30, 90 and 270 ng / ml could induce apoptosis of umbilical vein endothelial cells. When 30 ng / ml Apoptosis was induced, apoptosis was the strongest. After 2 hours, apoptosis of cells was observed With the induction of time, the rate of apoptosis increased gradually. Conclusion Staphylococcus aureus α-hemolysin can induce the apoptosis of human umbilical vein endothelial cells, and there is a significant time-dose relationship. This may be one of the important mechanisms that affect L-type vertical infection of S. aureus in fetal intrauterine growth.
