目的 利用Affymetrix基因芯片分析体外血管新生过程中内皮细胞的基因表达变化。方法 将人微血管内皮细胞包被于微载体 ,进行三维培养 ,以构建体外血管新生模型 ,然后在模型不同时间点提取内皮细胞总RNA ,采用Affymetrix芯片进行基因表达分析。结果 约 15 0 0个基因表达上调或下调 2倍以上 ,其表达呈现不同变化方式。结论 本研究从全基因组水平动态观察了体外血管新生过程中内皮细胞的基因表达变化 ,并与特定的血管新生形态学过程相联系。研究结果提供了丰富的血管新生分子信息 ,为进一步研究特定基因在血管新生中的作用提示了方向。
Objective To analyze the gene expression changes of endothelial cells during angiogenesis with Affymetrix microarray. Methods Human microvascular endothelial cells were coated on microcarriers and cultured in three dimensions to construct an in vitro model of angiogenesis. Total RNA was extracted from endothelial cells at different time points, and gene expression analysis was performed using Affymetrix microarray. Results about 1500 genes were up-regulated or down-regulated by more than 2 folds, and their expression showed different changes. Conclusion The present study dynamically observed the changes of the gene expression of endothelial cells in vitro during angiogenesis from the whole genome level and correlated with the specific angiogenesis process. The results provide a wealth of angiogenic molecular information, suggesting a direction for further studies on the role of specific genes in angiogenesis.
