目的:探讨葛根素对动脉粥样硬化(AS)兔主动脉血管平滑肌细胞(VSM C)核因子(NF)-κB表达的影响。方法:24只雄性日本大耳白兔,随机分为正常组、模型组、葛根素组。正常组饲喂普通饲料,模型组在普通饲料中加入1%胆固醇及0.02%蛋氨酸,葛根素组高脂饲料喂养的同时每天给予葛根素0.2g/kg。饲喂9周后,麻醉采血,取主动脉-80°C保存。测定各组兔血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(H DL-C);H E染色观察兔主动脉病理学改变;免疫组织化学检测兔主动脉NF-κB p65的表达。结果:与正常对照组比较,模型组TC、TG、LDL-C明显升高,H DL-C显著降低,差异有显著性或非常显著性意义(P<0.01或P<0.05);与模型组比较,葛根素组兔血清TC、TG、LDL-C明显降低,H DL-C显著升高,差异有显著性意义(P<0.05)。病理检验结果显示葛根素组AS病变显著延缓;葛根素组兔主动脉平滑肌细胞NF-κB p65表达明显减少,与模型组比较差异有非常显著性意义(P<0.01)。结论:葛根素可以改善血脂异常,抑制AS时血管壁慢性炎症反应。
Objective: To investigate the effect of puerarin on nuclear factor (NF) -κB expression in aortic vascular smooth muscle cells (VSMC) of atherosclerosis (AS) rabbits. Methods: Twenty-four male Japanese white rabbits were randomly divided into normal group, model group and puerarin group. The normal group was fed with normal diet. The model group was fed with 1% cholesterol and 0.02% methionine in the normal diet. The rats in the puerarin group were given puerarin 0.2 g / kg daily while they were fed with high fat diet. After 9 weeks of feeding, blood was collected by anesthesia and preserved in aorta at -80 ° C. The levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (H DL-C) in the rabbits in each group were measured. Pathological changes of the aorta were observed by HE staining Immunohistochemistry was used to detect the expression of NF-κB p65 in rabbit aorta. Results: Compared with the normal control group, TC, TG, LDL-C and HDL-C in model group were significantly lower than those in normal control group (P <0.01 or P <0.05) Compared with the control group, the levels of TC, TG and LDL-C in the puerarin group were significantly decreased, while the levels of H-DL-C in the puerarin group were significantly increased (P <0.05). The results of pathology showed that the pathological changes of AS in the puerarin group were significantly delayed. The expression of NF-κB p65 in the aortic smooth muscle cells of the puerarin group was significantly decreased compared with the model group (P <0.01). Conclusion: Puerarin can ameliorate dyslipidemia and inhibit the chronic inflammatory reaction of vascular wall in AS.
