目的研究合用雷公藤多苷片和阿托伐他汀之后,雷公藤多苷片对阿托伐他汀在大鼠体内药动学的影响。方法 12只雄性SD大鼠,随机分为A、B两组,其中A组为单独给药组,灌胃给药阿托伐他汀2 mg·kg~(-1),B组为联合用药组,灌胃给予雷公藤多苷片和阿托伐他汀,其给药剂量都为2 mg·kg~(-1)。在不同时间点取血,采用HPLC测定阿托伐他汀的血药浓度,计算并比较阿托伐他汀的药动学参数。结果与单独给药组相比,药物联用组阿托伐他汀的药动学参数发生了明显的变化,阿托伐他汀的最大血药浓度由(4.77±0.64)mg·L~(-1)上升到(7.79±0.61)mg·L~(-1),血浆曲线下面积由(12.82±3.50)mg·h·L~(-1)增加到(27.39±5.76)mg·h·L~(-1),同时达峰时间由(0.25±0.03)h延长到(0.52±0.07)h,半衰期由(2.39±0.19)h延长到(5.09±1.35)h,平均驻留时间由(2.93±0.23)h延至(4.36±0.44)h,说明阿托伐他汀的代谢可能受到了抑制。结论雷公藤多苷片和阿托伐他汀联用之后,会对阿托伐他汀的药动学产生一定程度的影响,导致达峰时间、半衰期延长,最大血药浓度增大,可能对阿托伐他汀钙的代谢具有抑制作用。本实验对雷公藤多苷片和阿托伐他汀联合用药具有一定的指导作用。
Objective To study the effect of tripterygium glycosides tablet on the pharmacokinetics of atorvastatin in rats after combination with tripterygium glycosides tablets and atorvastatin. Methods Twelve male Sprague-Dawley rats were randomly divided into A and B groups. Group A was treated with atorvastatin 2 mg · kg -1 by intragastric administration, while group B was treated with combination therapy , Intragastric administration of tripterygium glycosides tablets and atorvastatin, its administration dose is 2 mg · kg -1. Blood samples were taken at different time points. The plasma concentration of atorvastatin was determined by HPLC. The pharmacokinetic parameters of atorvastatin were calculated and compared. Results The pharmacokinetic parameters of atorvastatin in the combination group were significantly changed compared with those in the control group. The maximum plasma concentration of atorvastatin was (4.77 ± 0.64) mg · L -1 ) Increased to (7.79 ± 0.61) mg · L -1, the area under the plasma curve increased from (12.82 ± 3.50) mg · h · L -1 to (27.39 ± 5.76) mg · h · L -1, (-1), while the peak time prolonged from (0.25 ± 0.03) h to (0.52 ± 0.07) h, and the half life prolonged from (2.39 ± 0.19) h to (5.09 ± 1.35) h. The mean residence time increased from (2.93 ± 0.23) h to (4.36 ± 0.44) h, indicating that the metabolism of atorvastatin may be inhibited. Conclusions Tripterygium glycosides tablet combined with atorvastatin will exert a certain degree of influence on the pharmacokinetics of atorvastatin, resulting in peak time, prolonged half-life and increased maximum plasma concentration, Metabolism of statins of statins has an inhibitory effect. This experiment on Tripterygium glycosides tablets and atorvastatin combination therapy has a guiding role.
