Nitrogenase activity was measured using the acetylene reduction method in large soil circles (29 cm in diameter and 20 cm in depth) of Zea mays and sorghum. The enzyme activity and removal of roots from the soil, and then incubated with 1% O_2 overnight, and the method of determination of nitrogenase activity compared. However, nitrogenase activity was observed only after culturing and excising the root system. Not observed in plant analysis of large pots. This report compares nitrogenase activity of cultured and excised roots with that of corn grown in the Brazilian soil.
