目的 :一步法分析地高辛相对生物利用度和药物动力学参数。方法 :用NONMEM程序包及其新的POSTHOC子模块功能 ,估算地高辛相对生物利用度和药动学参数 ,并与经典的 3P87方法比较。结果 :两者的相对生物利用度一致 ;青年健康受试者口服地高辛的主要药动学参数典型值K10 (h-1)、Vc/F(L)和 β(h-1)分别为 0 .115 ,12 8和 0 .0 18,而老年病人的典型值K10 (h-1)、Vc/F(L)和 β(h-1)分别为 0 .0 40 3,76 .0和 0 .0 148。结论 :老年病人的主要药动学参数明显低于青年健康受试者 ,差异存在显著性。用NONMEM法在取样点数较少时可获得与经典法一致的个体药动学参数。
OBJECTIVE: To analyze the relative bioavailability and pharmacokinetic parameters of digoxin by one-step method. METHODS: The relative bioavailability and pharmacokinetic parameters of digoxin were estimated using the NONMEM package and its new POSTHOC submodule function and compared with the classical 3P87 method. Results: The relative bioavailability of the two drugs was the same. The main pharmacokinetic parameters of oral digoxin in young healthy subjects were K10 (h-1), Vc / F (L) and β (h-1) 0 .115, 12 8 and 0 .0 18, respectively. The typical values of K10 (h-1), Vc / F (L) and β (h-1) in elderly patients were 0.0403 and 76.0 0 .0 148. Conclusion: The main pharmacokinetic parameters of elderly patients were significantly lower than those of young healthy subjects, the difference was significant. With the NONMEM method can obtain the individual pharmacokinetic parameters consistent with the classical method when the sampling points are small.
