目的探讨Toll样受体2(TLR2)在局灶性脑缺血大鼠脑组织中的表达变化。方法应用免疫组织化学方法观察大脑中动脉阻断模型大鼠脑组织中TLR2蛋白表达水平,并实时定量反转录多聚酶链反应(real time PCR)方法检测大鼠局灶性缺血脑组织中TLR2 mRNA的相对表达水平。结果局灶性脑缺血后脑组织有TLR2阳性表达,对照组无TLR2阳性表达;局灶性缺血脑组织TLR2 mRNA表达水平较对照组明显上调(P<0.01)。结论大鼠局灶性脑缺血后脑组织中TLR2表达水平上调,提示TLR2可能参与了局灶性脑缺血的损伤过程。
Objective To investigate the expression of Toll like receptor 2 (TLR2) in brain tissue of focal cerebral ischemia rats. Methods Immunohistochemistry was used to observe the expression of TLR2 protein in the brain tissue of rats with middle cerebral artery occlusion (MCAO). The expression of TLR2 protein in focal ischemic brain tissue was detected by real time quantitative reverse transcription polymerase chain reaction The relative expression level of mRNA. Results There was TLR2 positive expression in brain tissue after focal cerebral ischemia, but no TLR2 expression in control group. The expression of TLR2 mRNA in focal cerebral ischemia group was significantly higher than that in control group (P <0.01). Conclusion The up-regulation of TLR2 expression in brain tissue after focal cerebral ischemia in rats suggests that TLR2 may be involved in the process of focal cerebral ischemia.
