目的:根据肾主骨生长发育理论,研究先天肾虚影响长骨早期发育的情况,论证补肾填精法的作用。方法:采用模拟地震振动、光电刺激复合应激恐吓孕鼠造成子代先天肾虚。将孕鼠分为正常、模型、补肾3组,补肾组灌服左归丸浓缩剂。动态检测出生后仔鼠20 d内平均身长、5周内股骨平均长度、Micro CT检测股骨皮质骨及松质骨微结构参数、第5周股骨钙、磷、羟脯氨酸含量。结果:模型组仔鼠平均身长、股骨平均长度显著降低(P<0.05或P<0.01);各时间点模型组皮质骨厚度、外径周长、皮质骨面积、皮质骨密度减少;股骨矿物质、基质胶原含量减少。上述指标与另两组比较差异显著(P<0.05)。结论:先天肾虚可以影响长骨早期生长发育,出现骨量减少,成骨活性降低,皮质骨发育不良。中药补肾填精治疗,可有效改善先天肾虚仔鼠长骨早期发育不良。
OBJECTIVE: To study the effect of innate kidney deficiency on the early development of long bones according to the theory of growth and development of the kidney, and to demonstrate the effect of the kidney-invigorating and filling method. Methods: Using simulated earthquake vibration, photoelectric stimulation of complex stress to intimidate pregnant rats caused offspring of kidney deficiency. The pregnant rats were divided into normal, model, kidney three groups, kidney group Zuo Gui Wan concentration group. The average length of the femur was measured within 20 days after birth, the average length of the femur within 5 weeks, the micro-structural parameters of cortical and cancellous femur measured by Micro-CT, and the content of calcium, phosphorus and hydroxyproline in the femur in the 5th week. Results: The average length of females and the average length of the femur in the model group were significantly decreased (P <0.05 or P <0.01). At each time point, the cortical bone thickness, outer diameter circumference, cortical bone area and cortical bone density decreased in the model group, , Matrix collagen content decreased. The above indexes were significantly different from the other two groups (P <0.05). CONCLUSION: Congenital kidney deficiency can affect the early growth and development of long bones, resulting in decreased bone mass, decreased osteogenic activity and cortical bone dysplasia. Chinese medicine kidney fill fine treatment, can effectively improve congenital Kidney Aberration of long bone early dysplasia.
