目的:观察热淋清颗粒对自身免疫性前列腺炎大鼠脾细胞增殖活性及前列腺组织炎性因子分泌的影响,进一步探讨其治疗慢性前列腺炎的可能作用机制。方法:采用大鼠前列腺蛋白提纯液辅以免疫佐剂造成实验性自身免疫性前列腺炎(EAP)大鼠模型,随机分为两组:治疗组给予热淋清颗粒原料药头花蓼浸膏2 g/kg;模型组给予相同体积的蒸馏水,另选一组正常组做对照。以上各组均灌胃给药,每天1次,连续给药30天。观察热淋清颗粒对各组大鼠脾细胞在刀豆蛋白A(ConA)、脂多糖(LPS)诱导下T、B淋巴细胞增殖活性的影响;采用放射免疫法检测分析热淋清颗粒对各组大鼠前列腺组织白细胞介素-1(IL-1)、IL-8、TNF-α及白介素-10(IL-10)等细胞因子分泌的影响。结果:热淋清颗粒能抑制脾淋巴细胞的增殖活性,抑制促炎细胞因子IL-1、IL-8及TNF-α的分泌,调节抗炎细胞因子IL-10的表达。结论:热淋清颗粒通过调节自身免疫性前列腺炎大鼠的免疫功能和促炎及抗炎因子的分泌而发挥治疗作用。
OBJECTIVE: To observe the effect of hot-linqing granules on the proliferation of spleen cells and the secretion of inflammatory cytokines in prostate tissue of rats with autoimmune prostatitis, and to further explore the possible mechanism of the treatment of chronic prostatitis. METHODS: A rat model of experimental autoimmune prostatitis (EAP) was induced by using protein purified from rat prostate and adjuvanted with adjuvant. The rats were randomly divided into two groups: g/kg; The model group was given the same volume of distilled water, and the other group was selected as the normal group. Each group was intragastrically administered once daily for 30 days. Observe the effect of heat-leaving granules on the proliferation activity of splenocytes of Conocan A (ConA) and lipopolysaccharide (LPS)-induced T lymphocytes in each group of rat spleen cells; The effects of cytokine secretion, such as interleukin-1 (IL-1), IL-8, TNF-α, and interleukin-10 (IL-10), in the prostate tissue of rats. Results: Hot-linqing granule could inhibit the proliferation of splenic lymphocytes, inhibit the secretion of pro-inflammatory cytokines IL-1, IL-8 and TNF-α, and regulate the expression of anti-inflammatory cytokine IL-10. Conclusion: Hot-linqing granules play a therapeutic role by regulating the immune function and secretion of proinflammatory and anti-inflammatory cytokines in rats with autoimmune prostatitis.
