目的制备盐酸利多卡因传递体溶剂,并对其理化性质进行评价。方法采用薄膜-超声法制备利多卡因传递体溶剂。用荧光显微镜和激光粒度分析仪观察监测盐酸利多卡因传递体的形态及粒度分布,以高效液相色谱法测定盐酸利多卡因含量来研究其稳定性。结果利多卡因传递体溶剂为近透明的胶体乳白色液体,荧光显微镜下可看到大量流动性的圆形或椭圆形药物载体并具有流动性,对盐酸利多卡因传递体溶剂进行测量其粒径为(81.1±3.1)nm;回收率为(100.2±0.35)%(n=3);包封率为(82.5±1.85)%(n=5),溶液应放置在4℃冰箱中保存。结论盐酸利多卡因传递体溶剂制备工艺可行,回收率和包封率良好,亦可满足分析实验要求。
Objective To prepare the solvent of lidocaine hydrochloride and to evaluate its physical and chemical properties. Methods The solvent of lidocaine transmitter was prepared by thin film - ultrasonic method. The morphology and particle size distribution of lidocaine hydrochloride were monitored by fluorescence microscopy and laser particle size analyzer. The content of lidocaine hydrochloride was determined by HPLC to study its stability. Results The lidocaine delivery solvent was a nearly transparent colloidal milky white liquid. A round or oval drug carrier with a large amount of fluidity could be seen under a fluorescence microscope and was fluid. The solvent of lidocaine hydrochloride delivery vehicle was measured for its particle size (81.1 ± 3.1) nm. The recovery was (100.2 ± 0.35)% (n = 3). The entrapment efficiency was (82.5 ± 1.85)% (n = 5). The solution should be stored in a refrigerator at 4 ℃. Conclusion The preparation method of lidocaine hydrochloride delivery solvent is feasible, the recovery rate and entrapment efficiency are good, and the requirements of analytical experiment can also be satisfied.
