为验证经高温加工过的鳖甲是否可以用 DNA分子标记鉴定 ,选取经高温高压蒸煮过的中华鳖鳖甲和骨进行 DNA提取 ,并用通用引物 L 10 91和 H1478扩增。结果表明经高温高压蒸煮过的鳖甲也可以提得较高质量的DNA,与从肌肉和药材鳖甲中提取的 DNA比较没有明显差异 ,并可用作模板进行 PCR扩增 ,得到约 40 0 bp基因片段 ;同时还讨论了 DNA提取和扩增过程中应注意的问题 ,为分子遗传标记技术广泛应用于同类药材的鉴定积累了基础资料。
In order to verify whether the heat-treated armor can be identified by DNA molecular markers, the extracted soft-shelled turtles and bones were used for DNA extraction and amplified with universal primers L10 91 and H1478. The results showed that the high-temperature and high-pressure cooking of the armor can also obtain higher quality DNA, and there is no significant difference compared with the DNA extracted from the muscle and medicinal armor, and can be used as a template for PCR amplification to obtain about 40 0 Bp gene fragment; also discussed in the DNA extraction and amplification process should pay attention to the problem, for molecular genetic marker technology is widely used in the identification of similar medicinal materials accumulated basic data.
