目的:建立以反相高效液相色谱法测定法半夏中γ-氨基丁酸含量的方法。方法:在碱性条件下,氨基酸与异硫氰酸苯酯反应,生成异硫氰酸苯-氨基酸的衍生物;采用Venusil-AA氨基酸分析柱(250mm×4.6mm,5μm)、二元梯度洗脱方式测定样品含量,流动相A为0.05mol.L-1醋酸钠溶液(pH6.5),流动相B为乙腈-水(80∶20),检测波长为248nm。结果:γ-氨基丁酸检测浓度在2.056~12.336μg·mL-1范围内与峰面积积分值呈较好的线性关系;加样回收率为99.0%,RSD=0.92(n=6)。结论:本方法简便、准确、重现性好,可用于法半夏的质量控制。
Objective: To establish a method for determination of γ-aminobutyric acid in Pinellia ternate by RP-HPLC. Methods: Under basic conditions, amino acids were reacted with phenyl isothiocyanate to generate benzene-amino acid derivatives; using Venusil-AA amino acid analysis column (250mm×4.6mm, 5μm), binary gradient wash The content of the sample was measured in the desorbing mode. The mobile phase A was 0.05 mol.L-1 sodium acetate solution (pH 6.5), the mobile phase B was acetonitrile-water (80:20), and the detection wavelength was 248 nm. Results: The γ-aminobutyric acid concentration in the range of 2.056~12.336μg·mL-1 showed a good linear relationship with the integral value of the peak area. The recovery rate of the sample was 99.0%, RSD=0.92 (n=6). Conclusion: This method is simple, accurate and reproducible. It can be used for the quality control of Pinellia ternata.
