初断乳雄性SD大鼠的肝匀浆以超速离心和柱层析法分离纯化肝刺激因子(HSS),观察其对肝癌细胞增殖、细胞表皮生长因子(EGF)受体表达及受体磷酸化的影响。结果表明,HSS具有明显的促肝癌细胞分裂增殖能力,提高细胞周期中S期细胞所占比例。HSS促肝癌细胞增殖作用与其促EGF受体表达有关,表现为:(1)HSS上调170kDEGF受体蛋白表达,此作用与EGF合用后明显加强,即是协同效应;(2)HSS上调EGF受体表达呈剂量-效应和时间-效应关系;(3)HSS可刺激EGF受体酪氨酸残基的磷酸化,调节受体活性。本研究结果提示,HSS可能是通过调节肝癌细胞EGF受体的表达及受体介导的信号传导通路而起到促增殖的作用。
The hepatic homogenates of primary male and female SD rats were subjected to ultracentrifugation and column chromatography to isolate and purify hepatic stimulating factor (HSS). The results of this study suggest that HSS may play a pro-proliferative role by regulating EGF receptor expression and receptor-mediated signaling pathways in hepatoma cells.
