目的:观察天智颗粒对慢性脑缺血大鼠海马CA1区小胶质细胞(microglia,MG)表达的影响,探讨天智颗粒的脑保护作用机制。方法:45只雄性SD大鼠,随机分为对照组、对照组、天智颗粒组。采用大鼠双侧颈总动脉永久阻断的方法制作慢性脑缺血模型。天智颗粒组在造模成功后给予天智颗粒(5 g·kg-1)ig 12周,采用Y迷宫、免疫组化染色和特异性尼氏染色方法观察比较各组大鼠学习记忆能力、OX42(小胶质细胞的标志)表达及海马CA1区神经元计数的差异。结果:与对照组相比,天智颗粒组学习记忆能力明显增加(P<0.01);对照组大鼠海马CA1区OX42少量表达,神经元数正常,对照组海马CA1区OX42表达明显增多,神经元数明显减少,天智颗粒组与对照组相比OX42表达明显减少,神经元数明显增加(P<0.01)。结论:天智颗粒能明显减少慢性脑缺血大鼠脑内OX42表达,抑制MG的过度激活,使海马区神经元死亡减少,具有脑保护作用。
Objective: To observe the effect of Tianzhi granule on microglia (MG) expression in hippocampal CA1 area of rats with chronic cerebral ischemia and to explore the mechanism of Tianzhi granule on brain protection. Methods: Forty-five male SD rats were randomly divided into control group, control group and Tianzhi granule group. Chronic cerebral ischemia models were made by permanent occlusion of bilateral common carotid arteries in rats. Tianzhi granule group was given Tianzhi granule (5 g · kg-1) ig for 12 weeks after successful modeling. The learning and memory abilities of rats in each group were observed by Y-maze, immunohistochemistry and specific Nissl staining. OX42 ( Microglia markers) and neuronal count in hippocampal CA1 region. Results: Compared with the control group, the learning and memory abilities of Tianzhi granule group were significantly increased (P <0.01); the expression of OX42 in hippocampal CA1 area of the control group was small, the number of neurons was normal, the expression of OX42 in hippocampal CA1 area was significantly increased, Compared with the control group, the expression of OX42 in Tianzhi granule group decreased obviously and the number of neurons increased obviously (P <0.01). Conclusion: Tianzhi Granule can significantly reduce the expression of OX42 in the brain of rats with chronic cerebral ischemia, inhibit the over activation of MG, reduce the death of neurons in the hippocampus, and protect the brain.
