目的:探讨清化颗粒对db/db小鼠肠道胰高血糖素样肽-1(GLP-1)合成和分泌的影响。方法:选取db/m小鼠为空白对照组,db/db糖尿病小鼠随机分为模型对照组和清化颗粒(低、中、高剂量)组,干预4周,观察小鼠糖代谢变化;ELISA法分析小鼠血清GLP-1的水平;qRT-PCR法检测小鼠回肠胰高血糖素原和激素原转化酶1/3(PC1/3)的mRNA水平;Western blot法检测小鼠回肠PC1/3的蛋白表达水平;组织病理学观察小鼠回肠组织形态变化。结果:与模型对照组比较,清化颗粒降低db/db糖尿病小鼠的空腹血糖(FBG)(P<0.01);提高血清GLP-1的浓度(P<0.01);上调小鼠回肠组织胰高血糖素原和PC1/3酶的mRNA水平(P<0.01);上调小鼠回肠组织PC1/3的蛋白表达水平(P<0.01);并具有剂量依赖效应。另外,清化颗粒明显改善db/db糖尿病小鼠回肠组织形态。结论:清化颗粒能够降低db/db糖尿病小鼠的血糖水平,提高血清GLP-1水平,上调胰高血糖素原和PC1/3的mRNA水平,促进肠道GLP-1的合成和分泌,改善回肠组织形态。
Objective: To investigate the effect of Qinghua granule on the synthesis and secretion of gut glucagon-like peptide-1 (GLP-1) in db / db mice. Methods: db / m mice were selected as the blank control group and db / db diabetic mice were randomly divided into model control group and Qinghua Granule (low, middle and high dose) groups. Interfered with for 4 weeks, the changes of glucose metabolism in mice were observed. The level of GLP-1 in mouse serum was analyzed by ELISA. The mRNA level of proglucagon and prohormone convertase 1/3 (PC1 / 3) was detected by qRT-PCR. The expression of PC1 / 3 protein expression levels; histopathological observation of mouse ileum tissue morphological changes. Results: Compared with the model control group, Qinghua Granules reduced fasting blood glucose (FBG) in db / db diabetic mice (P <0.01), increased serum GLP-1 concentration (P <0.01) (P <0.01). The protein level of PC1 / 3 in ileum of mice was increased (P <0.01), and the dose-dependent effect was observed. In addition, Qinghua Granules significantly improve ileal tissue morphology in db / db diabetic mice. Conclusion: Qinghua Granule can reduce blood glucose level, increase serum GLP-1 level, up-regulate mRNA level of PG and PC1 / 3 in db / db diabetic mice, promote the synthesis and secretion of GLP-1 in the intestine, improve Ileum tissue morphology.
