目的:研究风灵凝胶中秋水仙碱的体外透皮吸收和体外释放情况。方法:采用改良Franz扩散池装置,以离体SD大鼠皮肤、透析膜为体外渗透屏障,采用HPLC测定接收液中秋水仙碱的含量,计算秋水仙碱单位面积累积渗透量和释药量,比较透皮速率和释放速率。结果:风灵凝胶中秋水仙碱8 h的体外释放和经皮渗透行为均符合Higuchi动力学方程,体外释药量高于经皮渗透量,且释药速率高于透皮速率。结论:皮肤为风灵凝胶中秋水仙碱透皮吸收的主要屏障,该制剂能平稳的释放药物,具有长效缓释的特点。
Objective: To study the transdermal absorption and in vitro release of colchicine from Fengling Gel. METHODS: Using a modified Franz diffusion cell device, the skin and dialysis membranes of isolated SD rats were used as in vitro barriers. The content of colchicine in the receiving solution was determined by HPLC, and cumulative permeation volume and drug release amount per unit area of colchicine were calculated. Transdermal speed and release rate. RESULTS: The in vitro release and percutaneous permeation behavior of colchicine in Fengling Gel was consistent with the Higuchi kinetic equation. The drug release in vitro was higher than that of transdermal perfusion, and the drug release rate was higher than that of transdermal skin. Conclusion: The skin is the main barrier of transdermal absorption of colchicine in Fengling Gel. The preparation can release drugs smoothly and has the characteristics of long-term sustained release.
