目的:建立一种不需预先制备细胞单层的速成甲基纤维素半微量空斑技术。方法:以登革Ⅱ型病毒为代表,对速成法和常规法的实验条件、可靠性、敏感性、重复性等作系统研究。用方差分析和t检验对实验结果进行统计学处理。结果:速成法重复性好,简单快速,敏感稳定,速成法空斑平均水平高于常规法(α1>α2)。结论:速成甲基纤维素半微量空斑技术是一种可靠易行的病毒空斑新技术,可用于鉴定和滴定病毒,具有较大的实用和推广价值。
OBJECTIVE: To establish a rapid methylcellulose semi-micro plaque technique without pre-prepared cell monolayer. Methods: Dengue Ⅱ virus as the representative of the fast method and the conventional method of experimental conditions, reliability, sensitivity, repeatability and other systematic research. The variance analysis and t test were used to analyze the experimental results. Results: The fast method was good in repeatability, simple and rapid, sensitive and stable. The average level of rapid plaques was higher than that of the conventional method (α1> α2). Conclusion: The fast-acting methylcellulose semi-micro plaque technology is a reliable and easy new technology for virus plaque, which can be used to identify and titrate the virus, which is of great practical and popularization value.
